titleOfInvention | The use of pasteurella haemolytica glycoprotease in a process for recovering cells rich in o-glycosylated surface portions |
abstract | A method of recovering viable cells is described, the determinants of the surface proteins of which contain zones of carbohydrates with O glycosylation. The proteins are characterized by determinants which are sensitive at the substrate level to a neutralmetalloglycoprotease derived from P. haemolytica. The glycoprotease has a very limited specificity allowing it to perform the cleavage, on the surfaces of the cells, only of the parts with O glycosylation protein containing the determinants, while preserving the viability of the cells. Said method comprises: i) bringing the cells in solution into contact with affinity matrices which specifically bind to one or more determinants forming part of the substrates sensitive to the glycoprotease, the matrices having the possibility of binding to the determinants on cells; ii) separation of the cells to which the affinity matrices are linked from any material remaining in solution, the affinity matrices having a binding affinity for the determinants such that they remain linked to the determinants during the separation; iii) bringing the glycoprotease into contact with the separated cells in a concentration and for a sufficient time to effect the cleavage only of the parts of the substrate of the proteins having the determinants and linked by the affinity matrices, so as to thus release the affinity matrix cells. The released cells retain their viability due to the limited specificity of the glycoprotease which performs the cleavage only of the parts of the O glycosylation proteins presenting the determinants, and iv) the recovery of the cells thus released. |
