abstract | Detection of mutations in a lacZ gene comprising a lacZ operator sequence serving as a marker gene located in a plasmid, one or more copies of which are introduced into the DNA of a mammal or a mammalian cell. This detection method consists of isolating the DNA, cutting the plasmid from the DNA, recovering the DNA plasmid using solid particles to which a lacZ operator binding material is linked, releasing from new the plasmid of the particles after having isolated and then circularized it, to transform with the aid of the plasmid a bacterial host restriction of negative host, lacZ negative and galE negative, and to separate the transformants possessing a mutant gene of lacZ developing on a medium containing lactose, transformants having a non-mutant lacZ gene and which do not develop on such a medium. A transgenic mammalian model intended for mutation analysis has also been provided. |
