abstract | The invention relates to novel basic immunogenic polypeptides having epitopes for HCV (hepatitis C virus). The new basic immunogenic polypeptides are truncated polypeptides derived from the structural region of a human HCV isolate. Two new preferred basic immunogenic polypeptides are designated FGB1 and FGB2. The FGB1 and FGB2 polypeptides are believed to be derived from the vicinity of the N and C termini, respectively, of a putative protein (C) from HCV. In an ELISA, eight serum samples reacting to the anti-C100 HCV antibody (EIA, Ortho / Chiron), subjected to an assay by recombinant immunoblot I (RIBA I, Ortho / Chiron) and to neutralization (Neut., Abbot Labs.), Contained antibodies against the FGB1 and FGB2 polypeptides. In addition, the FGB1 and FGB2 and RIBA I polypeptides did not react with twelve serum samples which had reacted with EIA. In addition, two serum samples from a patient who was clinically diagnosed as not having hepatitis A and B, who did not react with EIA and RIBA I, reacted with FGB1 and / or FGB2. New polynucleotide sequences encoding the basic immunogenic polypeptides are also described. The present invention further comprises the application of these new sequences, of these polypeptides and antibodies developed against the new polypeptides in the detection of the hepatitis C virus, by immunoassays, diagnostics, the technique of enzymatic amplification of the genome and gene therapy. The invention also relates to novel immunogenic HCV polypeptides encoded in recombinant expression systems and clones, novel transformants, novel probes, novel methods of producing immunogenic HCV polypeptides, novel anti-idiotype antibodies, new non-coding polynucleotides and new kits. |
