Reagents and methods for identification of vaccines |
abstract |
Cells, serum or fractions thereof taken from exposed hosts, in particular characterized by an ability to protect against infections, constitute sorting agents making it possible to identify antigens intended for use in protective vaccines . Biological material taken from exposed native hosts can be validated in vivo in an unrelated host by their ability to destroy or neutralize the infectious agent. Validation is carried out by implantation of the infectious agent in a membrane envelope in an animal host to which the potential sorting reagent has been transmitted. Said potential reagent which successfully destroys or neutralizes the infectious agent can then be used to sort antigens produced by cDNA expression libraries or in extracts from the infectious agents in order to identify the components of effective vaccines. This process made it possible to identify potential immunogens against Dirophilaria immitis, in particular one of 39 kd. |
