abstract | The method for the detection of sulfidoleukotrienes sLT of the LTC4, LTD4 and LTE4 groups by a single enzyme-linked immunosorbent assay ELISA is based on the interaction occurring between one or more monoclonal anti-sLT antibodies and an sLT conjugated to a detection enzyme. A biological sample suspected of containing sLT is contacted with an anti-sLT monoclonal antibody bound to a vector. When the sLTs present in the sample have bound to the antibody, a conjugate composed of an sLT and a detection enzyme is added to the charged vector. Finally, the amount of the conjugate bound to the vector is evaluated, which amount is in inverse proportion to the sLT bound to the vector. This method is used for in vitro tests relating to the diagnosis of inflammatory diseases such as rheumatic diseases, immunodeficiencies, allergies or pseudo-allergies. If biological samples are primed with citokines, for example the initiators IL3, IL5 or GM-CSF, the sensitivity of the method can be increased. The assay is sensitive enough to detect sLTs produced by a few cells subjected to the reaction in tiny volumes of whole blood. |
