abstract | The invention relates to a process for the production of phenylthiocarbamyl (PTC) amino acids from phenylthiohyandotoin (PTH) or anilinothiozolinone (ATZ) amino acids. This process consists of a base-catalyzed ring opening of PTH or ATZ in the presence of a reducing agent. This process offers an alternative to the established Edman degradation reaction by aqueous acid conversion, in which amino acids of ATZ and PTC are converted to amino acids of PTH. The invention describes another method for producing reactive ATZ amino acids from PTH amino acids. These methods facilitate the analysis of proteins in low molar amounts, allowing the synthesis of amino acid derivatives thereof, which can be analyzed in amounts much smaller than the amounts required for traditional PTH amino acid analysis. |
