Assay methods and compositions for detecting and evaluating the intracellular transduction of an extracellular signal |
abstract |
Transcription-based techniques identifying extracellular signals that modulate the activity of cell surface proteins. The extracellular signals are identified by measuring the amount of transcription of a reporter gene in a recombinant cell expressing the cell surface protein and containing DNA coding for the reporter gene, the transcription of which is regulated, directly or indirectly, by said protein. cell surface. The techniques serve as a means of identifying potential pharmaceutical compounds which can be used to treat diseases due to their agonistic or antagonistic effects on the cell surface protein. Recombinant cells are also provided that express cell surface receptors and contain reporter gene structures which include transcriptional regulatory elements sensitive to the activity of cell surface receptors. |
