abstract | Derivatives of the growth hormone releasing factor GRF(1-29)NH2 and its analogs are prepared by reacting a substrate component of the formula GRF'-Met-Ser-X, wherein GRF' represents the initial sequence GRF(1-26) or its analogues, comprising fragments of GRF(n-26), in which n is between 1 and 8 and X represents an acyclic alpha-amino carboxylic acid residue having a side chain uncharged hydrophilic which exhibits at least the size of a methyl group, preferably Ala, Thr, Ser, Asn or Gln, with H-Arg-NH2 as the nucleophilic component, in the presence of an L-specific serine or thiolcarboxypeptidase enzyme obtained from yeast or of animal, plant or other microbial origin, preferably CPD-Y from yeast, in an aqueous solution or dispersion having a pH between 6 and 9 and, if necessary, coupling the desired fragment termination N (1-(n-1)) by chemical or enzymatic process. Also described are GRF-related peptides of the formula GRF"-Met-Ser-X, where GRF" represents the initial sequence GRF(1-26), in which 1 to 4 of the amino acid residues may be replaced or deleted, or their des-alpha-NH2 derivatives, X corresponding to the description above, and which constitute effective intermediate products. The method allows for selective transpeptidation, yielding satisfactory amounts of biologically effective GRF-related amides. |
