abstract Recombinant hosts containing and expressing various type II restriction endonuclease and / or modification methylase genes. The invention relates in particular to the cloned restriction endonucleases, NgoAIII and NgoAI, which recognize and cleave in or near the double-stranded DNA sequence, 5 'CCGCGG 3' and 5 'PuGCGCPy 3', respectively. The invention also relates to cloned modification methylase genes corresponding to said restriction endonucleases. In addition, the invention relates to a cloned modified methylase, M.NgoAII. One source of these enzymes is Neisseria gonorrhoeae, although other microorganisms can be used to isolate the isoschizomers of restriction endonucleases and the isoschizomers of modification methylases of the invention.
