Antibody molecule or molecule containing parts of antibody combining sites (catalytic molecule), which catalytically hydrolyzes a preselected amide or carboxylic acid ester bond from a reactive ligand, methods of making and use of the catalytic molecule, and cells producing these molecules. The molecules bind to a reactive ligand containing the bond to be hydrolyzed, and also to a haptenic ligand. The haptenic ligand is structurally analogous to the reactive ligand, and contains a tetrahedral carbon atom linked to a hydroxyl group, and to a saturated carbon atom at a position in the haptenic ligand, which corresponds to a position in the group. carbonyl and its bound heteroatom of the reactive ligand. The haptenic ligand also contains a group carrying an ionic charge in an aqueous solution at physiological pH values, which is not present at the corresponding position of the reactive ligand. The group carrying ionic charges is located in the hapten, at a distance not exceeding 7 Ã…ngstroms from the tetrahedral carbon atom. |
