abstract | A process for the preparation of interleukin 6 from transformed bacterial strains which comprises: a) the culture of a bacterial host transformed with an exspression vector coding for interleukin-6; b) bacterial lysis, isolation by centrifugation of inclusion bodies containing interleukin-6 and their denaturation with guanidine hydrochloride; c) dialysis of the denaturated proteins in the presence of a redox system, preferably reduced and oxidized glutathione; d) anion-exchange chromatography of the dialysate on DEAE-Sepharose, eluting with a pH adjusted saline water-glycerol solution gradient, selecting and collecting the fractions containing interleukin-6 at the desired purity. p |
