Polypeptides with prourokinase activity, plasmids coding therefor and process for their preparation and use |
abstract |
There are new polypeptides of the general formula that can be used as plasminogen activators Ser-X₁-X₂-rscuPA¹⁴³⁻⁴¹¹ (I) described in which X₁ and X₂ are defined amino acids or lower peptide bridges, X₂ can also be a simple bond and rscuPA¹⁴³⁻⁴¹¹ is the unglycosylated sequence range of amino acids 143 (Glu) to 411 (LeuOH) from the scu-PA 54 k. To obtain these compounds, new plasmids, which are described as such or in terms of their production, have been created, the expression of which in suitable bacterial hosts in high yields in each case provides an expression product which does not act as a plasminogen activator and whose protein-chemical conversion to the new single-chain plasminogen activators is valuable Compounds of formula I leads. |
