abstract | A method of isolating, cloning and maintaining large genomic fragments of chromosomal DNA. This method includes the steps of introducing a marker into the chromosomal DNA to be cloned, inserting double streaks into the chromosome and then immediately combining the cells containing the chromosome comprising the fragmented marker to another cell line capable of harboring extrachromosomal DNA elements, and then to isolate genomic fragments by separation of the extrachromosomal DNA from the chromosomal DNA of the other cell line. |
