abstract | A method of preparing an oil-in-water emulsion of a sparingly water-soluble drug, comprising dissolving the drug (e.g. amphotericin B) in a solution at high or low pH before the formation of the drug emulsion. The high pH solution is preferably a 0.5 M solution of sodium hydroxide and / or potassium hydroxide, and the low pH solution is preferably a 0.5 M solution of hydrochloric acid. The method comprises the steps of (a) dissolving the medicament in a solution of high or low pH, (b) adding the solution thus obtained to a preformed emulsion, (c) adding to the emulsion an amount of a acid, an appropriate base or buffer making it possible to neutralize at least the product of stage (b), and (d) when an acid or a base is added in stage (c), optionally adding a sufficient buffer allowing the pH of the product of step (c) to be adjusted to the desired value. A drug emulsion produced by the method was also produced, in which the drug was first associated with the oil droplets. |
