abstract | The DNA multiplex sequencing process includes blot-drying electro-drying of DNA fragments terminated by a specific base and which have been resolved by gel electrophoresis, on the surface of a neutral non-aromatic polymer microporous membrane having low background fluorescence and the surface of which has been modified to contain amino groups. Polypropylene membranes are preferred and the introduction of amino groups is carried out by subjecting the membrane to a plasma discharge at high frequency or at microwave frequency in the presence of an aminating agent preferably ammonia. The membrane, which contains DNA fragments physically adsorbed on its surface after electro-drying, is then dried with crosslinking means such as UV radiation or an atomization of glutaraldehyde to chemically bind the DNA fragments on the membrane by through the amino groups contained on its surface. The DNA fragments chemically linked to the surface are subjected to a hybridization probe with a labeled probe specific to the sequence of the DNA fragments. Labeling can be carried out either using fluorophores or using radioisotopes. The labeled probes hybridized to the target DNA fragments are detected and read by detection of fluorescence induced by laser or autoradiograms. The use of amino membranes with low background fluorescence makes it possible to use fluorescence detection and reading even when the available quantity of DNA to be sequenced is small. DNA bound to membranes can be detected by probe many times. |
