abstract | The present invention improves the resolution of a confocal and scanning microscope by performing optical signal processing prior to detection. This processing is carried out by an element (7, 39, 42) having a weighting, either in phase, or in amplitude, or even both. The microscope can be a transmission microscope, a reflection microscope, or a fluorescent microscope. If it is a transmission microscope, a laser (1) illuminates a small part of the sample (3). The light from this small part is focused on an optical processing element (7). The output signal from this element is detected by a detector (1) via a perforation (9) made in a plate (10). The phase weighting of the element (7) can be carried out by a rectangular network (15), and the amplitude weighting, by a sinusoidal variation of the absorption. A function m(x, y) of the optical weighting is cos(2piNx/lambda).cos(2piNy/lambda) where N is the numerical aperture of the lenses (2, 6) and lambda is the wavelength. Another n(x, y) optical weighting function is sign(cos(2piNx/lambda)).sign(cos(2piNy/lambda)). |
