The present invention relates to pharmaceutical compositions and methods of producing compositions containing liposomes and having high uptake characteristics. This capture can be greater than that which one supposes by applying the relation defined by the Henderson-Hasselbach equation. The invention also relates to liposomal compositions in which the liposome partly comprises a membrane stabilizing component, for example cholesterol, which has favorable characteristics for preventing rapid release of a pharmaceutical agent selected from the group comprising quinine. , quinidine, and diphenhydramine after formulation in liposomes. The invention also relates to novel liposomal compositions comprising the bronchodilators, namely metaproterenol, isoproterenol and terbutaline. The invention also relates to a minimum buffer capacity required to obtain liposomal encapsulation of pharmaceutical agents with maintenance of a large part of the initial pH gradient. |
