abstract The invention relates to a method for the construction of plasmids ensuring the insertion of a nucleotide sequence into a chromosome and the simple separation of clones containing the transformed chromosome using known genetic manipulation methods, comprising inserting a copy of a chromosomal segment corresponding to a chromosomal segment of the host cell between the transcriptional regulatory sequences, capable of expressing a foreign gene in a plasmid containing transcriptional regulatory sequences capable of expressing a foreign gene as well as a foreign gene in a plasmid containing transcriptional regulatory sequences capable of expressing a foreign gene, and a resistance gene (R2) expressed independently of said regulatory sequences; a resistance gene without its own promoter (R1) different from (R2) is then inserted downstream, then, finally, a sequence containing at least one transcription terminator in the inserted copy of the chromosomal segment.
