abstract |
The methods described for isolating the thymidine kinase coding DNA from a herpes virus use degenerate primers based on regions of a relatively conserved amino acid sequence in thymidine kinase proteins herpes virus to initiate a polymerase chain reaction that produces large amounts of DNA encoding thymidine kinase. The methods are illustrated in the isolation of the feline herpes virus thymidine kinase gene which can be used to construct recombinant thymidine kinase negative feline herpes viruses for the purpose of constructing live and expression vectors. In addition, the regulatory elements of the feline herpes virus thymidine kinase gene are useful in the construction of recombinant DNA vectors. |
