abstract | The invention comprises a method and an apparatus for the analysis in sequence of single and double-stranded nucleic acids on solid phase supports, according to which the terminations of nucleic acids intended to be sequenced using units are lengthened. modified nucleic acids, which serve as units for anchoring to a water-insoluble and solid polymeric support. In the preferred embodiment, the modified nucleotides to be attached to a chain of nucleic acids by catalyzed chemical or enzymatic reactions are in particular 5-bromo-2'-deoxyuridylate or in general other nucleotides which are halogenated or suitably substituted differently at the level of the basic or sugar fractions, or of streptavidin. The nucleic acids that can be immobilized in this way are either DNA or single-stranded RNA chains. The water-insoluble and solid supports are polymers such as cellulose, sepharose or sephadex, as well as inorganic supports, in particular supports whose skeleton structure consists mainly of silicon and oxygen atoms. The solid supports which are insoluble in water comprise effector groups, they may in particular contain immobilized antibodies specific for 5-bromo-2'-deoxyuridylate or for other appropriate units capable of selective interaction with other halogenated nucleotides or nucleotides suitably substituted at the base or sugar moieties. The strands of nucleic acid to be sequenced are attached to the polymer support by interactions of the modified nucleotide units with the effector groups immobilized on the support, in particular by interactions of receptor ligands. An apparatus has been developed for carrying out the process. |
