abstract | This invention relates to genes called Rpt-1 (regulatory protein T lymphocyte-1) expressed at higher levels by resting CD4 + T helper / inducer cells compared to activated CD4 + cells. The invention also relates to proteins encoded by such genes, called rpt-1 proteins, which regulate gene expression caused by the activator region of the alpha-chain gene of the interleukin-2 receptor (IL-2r) or by activator region of the repeated terminal sequence of human lymphotropic retroviruses such as human immunodeficiency virus type 1 (HIV-1), human T cell leukemia virus (HTLV) -I, and in particular HTLV-II. The rpt-1 proteins negatively regulate gene expression driven by the activator of the HIV-1 repeat terminal sequence. The proteins and nucleic acids of the invention are useful in the diagnosis and therapy of immune disorders such as AIDS. An Rpt-1 gene and its encoded intracellular protein of molecular weight of approximately 41,000 daltons are described in a specific example of the present invention. The rpt-1 protein is presented as being selectively expressed by activated CD4 + T cells, and negatively regulating the gene expression of IL-2r and HIV-1. |
