abstract | This invention relates to an enzyme assay of biological substances utilizing hydrogen peroxide as the reaction product in a NAD(P)-NAD(P)H system, which comprises first adding isocitric acid, metallic ions and isocitric acid dehydrogenase to a specimen to thereby consume intrinsic substances inhibiting the assay and at the same time reproduce decreased NAD(P)H, then adding a chelating agent thereto to deactivate the isocitric acid dehydrogenase, further adding simultaneously or thereafter an enzyme or a substrate capable of producing hydrogen peroxide as a reaction product thereto, and measuring produced hydrogen peroxide. Since the intrinsic substances which inhibit the assay are removed, the assay of the present invention enables the biological substances to be accurately determined based on the measurement of the produced hydrogen peroxide without being affected by said intrinsic substances. |
