A method of expressing a protein in Aspergillus comprising transforming a strain of Aspergillus with a vector system encoding a promoter having upstream enhancer sequences derived from an A. niger amylase, a marker suitable for selection of transformants and a DNA sequence encoding the derived protein. This process allows the industrial production of many different polypeptides and proteins in Aspergillus, preferably A. niger. These products are for example chymosin or prochymosin and other rennets, proteases, lipases and amylases. Another object of the invention is an efficient promoter intended to express a protein in Aspergillus, preferably Aspergillus niger, and which is derived from a gene encoding the amylase of A. niger. The amylases of A. niger are the neutral and acid-stable alpha-amylases and a new amylase heretofore described as amylase XA and designated by this name. Another object of the invention is the new amylase from amylase XA of A. niger. |
