abstract | Live vaccines and methods for preparing live vaccines protect a host from pathogenic microorganisms. These vaccines are prepared by introducing at least one modification to a gene involved in at least one, normally at least two, biosynthetic pathways requiring the production of substances probably unavailable in the host capable of catching the disease. The modification causes a genetic change which cannot be eliminated in a single step, for example by erasure or by inversion of polynucleotides. When it is the aro genes that undergo such a change, the auxotrophic mutants obtained require aromatic amino acids, p-aminobenzoic acid and 2,3-dihydroxybenzoic acid or an extremely concentrated source of absorbable iron. Auxotrophic mutations show substantially reduced or no virulence but retain the desired immunogenicity in order to trigger the immunogenic response. Several techniques can be used to achieve the desired change. Salmonella typhimurium strain SL1479 was deposited with ATCC on September 7, 1982 and received registration number 39183; Salmonella dublin strain SL1438 was deposited on September 7, 1982 with ATCC and received registration number 39184. Salmonella typhi strain 531 ty was deposited with ATCC on November 21, 1984 and received number registration 39926. Shigella flexneri strain SFL114 was deposited with ATCC on March 14, 1988 and received registration number 53755. |
