abstract | Polyanhydrides having a uniform distribution of aromatic and alkyl residues are prepared by fusion-polycondensation or by solution polymerization of p-carboxyphenoxyalkanoic acids or p-carboxyphenylalkanoic acids. These polymers are soluble in common organic solvents and have low melting points, generally 40 to 100 ° C. Polyanhydrides are particularly well suited for the formation of a bioerosion matrix in devices for the regulated supply of bioactive compounds. A polymer matrix formed by the process described here degrades uniformly during drug release, preventing the complete channeling of the bioactive compound into the environment, and eliminating the problem of the presence of the polymer matrix long after the drug is released. The polymer exhibits zero order cynetic degradation profiles over various time periods (a few days to several months) at a rate useful for regulated drug delivery. In addition, a desired degradation rate can be obtained by choosing the appropriate length of the half or aliphatic fraction. |
