abstract Improved methods for preparing variants of human t-PA (tissue plasminogen activator) proteins with pharmacokinetic properties compared to natural t-PA. An illustration of such a variant, free of amino acids corresponding to amino acids ranging from 1 to 44 of t-PA, shows a half life of plasma greater than about 15 times half life of plasma of natural t-PA , as well as an elimination rate less than about 1/10 of the elimination rate of natural t-PA. Also disclosed are improved methods for treating vascular diseases using pharmaceutical compositions containing therapeutically effective amounts of said t-PA variants with pharmaceutically acceptable diluents or excipients.
