abstract | LEU3, a leucine-specific regulatory locus, encodes a protein for control of RNA levels of a group of leucine-specific genes. The protein encoded by LEU3 binds to a decanucleotide core sequence in the promoter regions of LEU1, LEU2, LEU4, ILV2, and, by implication, ILV5. The largely conserved decanucleotide core sequence shared by the binding sites in these genes is C C G G N N C C G G. DNA sequence analysis of a clone of LEU3 shows that it contains an open reading frame of 886 amino acids. A striking feature of the LEU3 protein is a cluster of acidic amino acids (19 out of 20) that are located in the C-terminal half of the coding region. The protein also has a repeated cysteine motif and a region of partial homology to MAT alpha 2 and the homeo box domain. LEU3, or a portion of LEU3 consisting of the N-terminal 160 amino acids and the C-terminal 112 amino acids, and the corresponding LEU3 decanucleotide binding sequences are useful in the induction or regulation of protein synthesis including not only expression of LEU1, LEU2, and LEU4, but also any gene wherein the DNA binding sequences are incorporated into the 5' region of the gene. |
