abstract |
Preparation and in vivo testing of surface coatings and matrix materials which, when containing or applied to carriers of drugs and diagnostic agents, are administered to provide effective vascular access, recognition by the support of determining substances present on a normal or locally affected endothelium, and bring about the following in vivo effects: 1) rapid, partial or total envelopment of the support for the drug (or diagnostic agent); 2) sequestration of the support and rapid protection (2 minutes) of the encapsulated agent of blood vascular purification when the endothelial pocket which envelops the support is still invaginating the vascular compartment; 3) acceleration of the transport of the support through the vascular endothelium or the subendothelial structures towards the interstice; and 4) improving the migration efficiency of the drug (or diagnostic agent) across endothelial or subendothelial barriers, thereby reducing the total dose of drug required to achieve the desired effect compared to the corresponding dose in usual agents. Analogous tissue absorption is obtained for transepithelial migration in the lungs, bladder and intestines. |
