abstract | The invention provides a method and kit for the determination of nNAD(P)H in a sample, e.g. a clinical sample, which comprises reducing npyruvic acid or a salt thereof with NAD(P)H in the presence of lactate ndehydrogenase to form lactic acid, oxidizing the lactic acid in the presence nof lactate oxidase to form hydrogen peroxide and determining the hydrogen nperoxide by reaction with peroxidase in the presence of a chromogen nunsusceptible to NAD(P)H. n The method is especially useful in determination of analytes, e.g. bile nacid, phosphohexose isomerase, in serum containing lactic acid, in which ncase the sample is initially reacted with lactate oxidase to convert the nlactic acid to pyruvic acid and H₂O₂, which is then decomposed, the pyruvic nacid produced in that case providing at least a proportion of the pyruvic acid nwhich is subsequently converted back to lactic acid. |
