abstract |
The baculovirus-insect cell vector system is useful as a high efficiency eukaryotic cloning and expression method for the production of the recombinant proteins. A major application of this system is in the development of rDNA vaccines for important human and animal diseases. This system is also useful to express the circumsporozoite antigen of a parasite that causes malaria in humans, Plasmodium falciparum . The resulting recombinant protein consists of full-length circumsporozoite antigen. The results indicate high levels of expression into recombinant products which closely resemble the authentic product. Furthermore, it has been demonstrated to effect purification of the recombinant product to near homogeneity. The expression of this gene by the baculorvirus-insect cell vector system yields these desirable results because this gene is normally expressed by insects. Recombinant circumsporozoite proteins produced in this manner have application as ingredients for sub-unit vaccines to protect against malaria, and as an ingredient for malaria diagnosis. |
