abstract | It is now known that thrombin is the mediator of a plurality of powerful biological effects concerning cells carrying receptors with high affinity for thrombin. These effects depend, at least in part, on receptor occupancy signals generated by the interaction between thrombin and the receptor with high affinity for thrombin. The inventors formulated synthetic thrombin derivatives capable of selectively stimulating or inhibiting the occupation signals of the thrombin receptors. Thrombin stimulating derivatives bind to thrombin receptors located on the surface of cells and stimulate DNA synthesis in cells treated with non-mitotic concentrations of alpha-thrombin or phorbol myristate acetate. These peptides, which have a thrombin receptor binding domain and an amino acid segment having a sequence common to several serine proteases, thus seem to generate mitotic signals dependent on the occupation of the receptors. The inhibitory derivatives, which do not have amino acid sequences conserving serine esterase, bind to thrombin receptors without generating mitotic signals depending on the occupation of the receptors. The invention relates to these peptides and methods of using them to promote cell growth and wound healing or to inhibit scar formation, tissue adhesion, tumor metastasis and angiogenesis. |
