abstract A biological substance is determined using a label comprising a lanthanide or other metal ion coupled to the biological substance using a chelate-forming compound. The chelated complex thus formed is dissociated by reducing the ambient pH to a level low enough to release the metal ion, which is then subjected to chelation with a separate chelating chromophore, accompanied by an increase in pH to a value. close to the optimum for determining the metal chelation chromophore by fluorescence spectroscopy, luminometry, colorimetry or other similar techniques.
