This invention relates to a method for preparation of high purity diglyceride with high yield in which esterification reaction between saturated or unsaturated fatty acids with 4 to 22 carbon atoms or lower alcohol ester thereof and glycerin is carried out in the presence of 1,3 position selective immobilized or endo-lipase with removing water or lower alcohol generated from the reaction out of system as much as possible to increase ester synthesis yield and to decrease the amount of monoglyceride. 1.5 mol or more fatty acids mentioned above are suitably used to 1 mol glycerin, more preferably 1.5 to 2.2 mol fatty acids are used. In accordance with the present invention, diglycerides can be obtained with high purity and high yield which can not be obtained by conventional methods. |
