titleOfInvention METHOD FOR MONITORING THE SYNTHESIS OF A LINEAR COMBINATION OF AMINO ACID RESIDUES.
abstract The control process described below applies particularly (but not exclusively) to the synthesis of peptides in solid phase. In one aspect of the present invention, the synthesis begins with an amino acid residue protected by an N-alpha-amino protecting group and consists in: (a) removing the N-alpha-amino protecting group in order to obtain a N-alpha-amino group, (b) add, using a peptide bond, an amino acid residue protected by an N-alpha-amino protective group to the N-alpha-amino group obtained in (a) , using a protected reactive amino acid derivative and if necessary a catalyst, and (c) repeat steps (a) and (b) until the desired linear combination is obtained. The reaction system comprises a 3-hydroxy-1,2,3-benzotriazine-4 (3H) -one group or a derivative of said group. The color of the reaction system or of a component of said system is checked during the system. The solvent is preferably a polar aprotic solvent, for example a dimethylformamide. When the synthesis is a solid phase synthesis, the solid phase is preferably a polyamide. The present invention also describes an apparatus for carrying out said methods. In a preferred embodiment, said apparatus comprises (i) a column intended to receive particles comprising a polyamide resin on which the solid phase synthesis is carried out, (ii) a light source placed on one of the sides of said column , (iii) a focusing device placed adjacent to said column and opposite to said light source, (iv) a photometer placed so as to receive the light focused by said focusing device, and (v) a programmed microprocessor to react according to the output of said photodetector, so as to control the synthesis procedure.
