abstract | Plasmid allowing the expression of a foreign polypeptide which, when it is introduced into an appropriate E. coli cell carrying a heat-inducible RecA protease gene and when it is cultivated under the appropriate conditions, expresses d a foreign gene inserted into the plasmid and the production of the foreign polypeptide. The plasmid is a double-stranded DNA molecule having an order of 5 'to 3' as follows: a DNA sequence which includes a heat-inducible promoter, with its associated ribosome binding site, which promoter is derived from a bacterial plasmid and normally causes the expression of a bacteriocin selected from the group consisting of colicin and the coded peptide and by the col gene; a DNA sequence which codes for an ATG initiation codon; a DNA sequence that codes for the foreign polypeptide; a DNA sequence which is a gene facilitating high copy number replication of the plasmid; a DNA sequence which is a gene associated with a phenotypic trait which can be chosen or identified and which manifests itself when the plasmid is present in the host cell; and a DNA sequence containing an origin of replication from a bacterial plasmid capable of autonomous replication in the host cell. |
