abstract | Efficient precipitation and extraction of fibrinogen and fibronectin proteins from blood plasma fractions, especially cryoprecipitate, leaving large amounts of blood coagulation factor VIII in the supernatant. This is achieved by the addition of at least 0.15 mg, preferably 0.3-0.9 mg, of a sulfated polysaccharide, including heparin, per ml of buffered plasma fraction solution while keeping the temperature of the solution at a level above 15oC, preferably between 20 and 35oC, during the extraction of fibrinogen or fibronectin. Freeze-dried factor VIII preparations prepared from the factor VIII-rich supernatant are suitable for heat treatment, e.g. at 70oC for 24 hours during inactivating the viruses generated in the blood, without significant production of insoluble denatured protein by-products. |
