abstract A method of sequentially eliminating one or more residual amino acids from an analog of a eukaryotic polypeptide synthesized in a foreign host includes contacting the eukaryotic polypeptide analog with aminopeptide under suitable conditions allowing elimination of N-terminal residual amino acids, while the polypeptide analog contains a residual amino acid or sequence thereof that blocks the action of an aminopeptide located elsewhere than the N-terminus of the polypeptide analog. A preferred embodiment involves the removal of residual N-terminal methionine and adjacent residual leucine from a growth hormone analog produced in bacteria by contacting the growth hormone analog with aeromonas aminopeptidase under appropriate conditions allowing the elimination of N-terminal methionine and residual leucine. The invention also relates to the eukaryotic polypeptide analogs obtained by this method.
