@ A method for transforming thermophilic microorganisms. A method for transforming thermophilic microorganisms is disclosed comprising: (a) forming a protoplast from a thermophilic microorganism; (b) introducing a plasmid chimera into the protoplast; and (c) causing regeneration of the cell wall of the protoplast. A novel regeneration medium is employed in the transformation method. This method involves the use of novel plasmid chimera. The plasmid chimeras include a newly discovered thermostable promoter. A method of isolating promoters capable of functioning at elevated temperatures is included. A method for selecting thermostable variants of gene products of cloned genes in thermophilic microorganisms is also disclosed. |
