abstract A method of concentrating and measuring the amount of ncells in a sample is described. A sample containing the nplurality of cells is passed upwardly or laterally through a nseries of membrane filters and concentrated on the filter. The nfilter membrane containing the concentrated cell is then nsequentially treated with lysing agent to release the nadenosine triphosphate from the organism and a luminescent nreagent to react with the released adenosine triphosphate. A nluminescent response is thereby produced directly on the nfilter membrane and this luminescent response is measured nwith a luminometer. Not only does this method provide an naccurate way of detecting very minute quantities of cells but it nprovides a relatively quick and easy method of concentrating nand measuring cells in large volumes of biological or nindustrial samples which contain low levels of organisms. nThis method can also be used to identify and measure sources nof adenosine triphosphate. Another aspect of the disclosed nmethod is the utilization of a series of filters, each of which ntraps and concentrates cells of different size. For example, a nsample containing a plurality of cell types can be passed nthrough a series of filter membranes, with different pore size neach of which allows passage of all but one unicellular norganism, resulting in a concentration of different cells on ndifferent filter membranes. The filter membranes can then be nprocessed and measured on the luminometer as described nabove.
