FRAGMENT OF DNA MARKED WITH AT LEAST ONE OF THEIR ENDS BY MODIFIED RIBONUCLEOTIDES RECOGNIZABLE BY AFFINOUS MOLECULES AND METHOD FOR PERFORMING AN ANALYSIS OF SUCH DNA FRAGMENTS. |
abstract |
Method for analyzing DNA sequences. It consists in attaching to one of the ends of this DNA a ribonucleotide carrying a molecule covalently attached and carrying a modification group couplable with an enzyme, allowing their subsequent visualization in the presence of a terminal DNA transferase, to treat this DNA with separate chemical agents making it possible to effect differential cleavages within this DNA at the level of bases, also of respective natures, to collect and separate the DNA fragments carrying the groups which can be viewed in a system enabling them to be sorted by size order, and to determine the unmodified terminal nucleotides of these DNA fragments, having regard to the nature of the chemical agents used for cleavage at their respective levels. |
