titleOfInvention Linear kinetic determination of bicarbonate in body fluids
abstract A reagent and process is provided for the linear determination of bicarbonate concentration in a body fluid. A reaction mixture is formed by combining predetermined volumes of body fluid and a reagent containing (i) non-rate-limiting amounts of phosphoenolpyruvate and the reduced form of nicotinamide adenine dinucleotide, (ii) phosphoenolpyruvate carboxylase and malate dehydrogenase, (iii) inorganic pyrophosphate, and preferably (iv) oxamate. The decrease in absorbance of the mixture is measured at a wavelength in the range of 320 nm to 400 nm at a fixed-time interval while maintaining the mixture at a substantially constant pH and temperature, and the measured absorbance decrease is related to the decrease in absorbance (measured under the same conditions) of a mixture formed by combining the reagent with a solution of known bicarbonate concentration.
