titleOfInvention | Pyrimido(4,5-b)(1,4)thiazines and process for their preparation |
abstract | The present invention relates to 7H-pyrimido [4,5b] [1,4] thiazines of the formula I, <IMAGE>, which are obtained by cyclization from the corresponding pyrimidines and α-haloketones. These compounds inhibit the de novo biosynthesis of folic acid in microorganisms and thereby inhibit their growth. They can therefore be used in pharmacological, clinical and diagnostic tests that affect the properties of bacteria. As bacteriostatics, these compounds reduce the rate of growth of microorganisms at a concentration of> = 100 µmol / l. In addition, combinations of these substances with sulfonamides and / or dihydrofolic acid reductase inhibitors of the 2,4-diamino-5- (3 ', 4', 5'-trimethoxybenzyl) pyrimidine (TMP) type act synergistically by sequentially blocking two enzymes, which is due to bacterial multiplication kinetics can be shown. This mutual potentiation of the action of individual inhibitors delays the development of resistance in microorganisms and expands the spectrum of the antimicrobial action of this combination, since organisms that are only slightly sensitive to the individual substances can be strongly inhibited in their growth by the combination. |
