abstract A prostate antigen different from prostatic acid phosphatase has been detected in normal, benign hypertrophic and malignant prostate tissue, but not in other human tissue. Prostate antigen was purified to homogeneity from prostate tissue by precipitation of ammonium sulphate, anion exchange chromatography DEAE-BioGel A, molecular sieves on Sephadex G-100 and Sephadex G-75, and polyacrylamide gel electrophoresis. The purified prostate antigen presents a single protein band by analytical electrophoresis on polyacrylamide gel and by isoelectric focusing. The molecular weight of the purified antigen was estimated by filtration on Sephadex G-75 gel at 33000 and by electrophoresis on polyacrylamide-sodium sulfate dodecyle at 34000 without subunit. The prostate antigen had an isoelectric point of 6.9.
