Polynucleotides are produced in a semi-automated process using a solid, polymeric support to which an initial nucleotide unit is initially chemically attached. In subsequent chain extension steps, the polymer-nucleoside is reacted with a predetermined 5'-blocked nucleoside complex and blocked phosphodichloridite (which may be prereacted), following suitable unblocking of the 5'-position of the polymer- nucleotide complex. A polynucleotide of any desired chain length and chain sequence is builtup sequentially in this way. Oxidation of all phosphite linking groups to phosphate is conducted in a single step after chain extension has been completed. |
