abstract |
In a controlled protein fractionation process a cold protein-containing solution is circulated in contact with one side of a membrane and the temperature is controlled by circulating a solution of a protein precipitant in contact with the opposite side of the membrane. The flow is increased and a reaction pressure is exerted on the precipitant solution which forces its penetration into the membrane, its mixture with the protein solution and precipitates the proteins in fine particles along an inner membrane wall. The precipitated proteins can be concentrated and separated by centrifugation or diafiltration. The membrane is preferably in the form of a plurality of hollow fibers. Controlling process variables increases yield and purity, decreases processing time and reduces the degree of denaturization. The method is especially applicable to the Cohn cold alcohol fractionation process. |
