abstract | Certain peptides administered during chronic morphine therapy have been shown to be effective in preventing the development of physical tolerance and dependence. Particularly effective peptides are prolyl-leucyle-glycinamide, cyclo (leucyleglycle), and the protected peptide- N-carbobenzoxy-prolyl-D-leucyle, which, injected daily into mice receiving chronic morphine treatment, have prevented development of physical dependence, which has been measured by changes in temperature and body weight, either during sudden cessation or while taking naloxone. However, the injection of a peptide only on the last day of morphine treatment did not alter the obvious signs of stopping. Daily injection of a peptide has also been shown to block the development of tolerance to the analgesic and hypothermic effects of subsequent doses provoking morphine. Treatment with peptides did not alter the acute effects of morphine on analgesia or body temperature. No effect on the memory was noticed as it could be evaluated in an exercise of test with passive cancellation. |
