abstract Synthetic DNA coding for a mammalian polypeptide, ne.g. somatostatin, is prepared in which at least a majority of nthe codons are those preferred for the expression of nmicrobial genomes. The DNA is prepared by joining together ntwo appropriately coded series of fragments by hydrogen nbonding and then completing the two strands by ligation, the nfragments lacking complementarity other than in the correct nsequence. n Such synthetic DNA having a restriction endonuclease nsite at each end of the gene can be introduced into a nmicrobial cloning vehicle which can then be used for ntransformation of bacterial hosts.
