Cysteine-modified monospecific and bispecific molecules containing EGFR and / or c-Met FN3 domain, including one or more free cysteine amino acids, are obtained by mutagenesis of the nucleic acid sequence of the original molecule and replacing one or more amino acid residues with cysteine to encode monospecific or bispecific molecules containing a cysteine-modified FN3 domain; expression of molecules containing a cysteine-modified FN3 domain; and isolating a molecule containing a cysteine-modified FN3 domain, isolated monospecific or bispecific molecules containing a cysteine-modified FN3 domain can be covalently attached to a detectable label or drug molecule and be used therapeutically. |
