abstract This invention relates to highly active cyclic dinucleotide (CDN) immune stimulants that activate DC through a newly discovered cytoplasmic receptor, known as STING (Stimulator of Interferon Genes - an interferon gene stimulator). In particular, the CDN of this invention is presented as a composition containing one or more cyclic purine dinucleotides that induce human STING-dependent production of interferon type I, where the purine cyclic dinucleotides present in the composition are 2'-fluoro-substituted, bis-3 ' 5 'CDN and most preferably one or more 2', 2 "'- diF-Rp, Rp, bis-3', 5'CDN.
