abstract According to the present invention, inter alia, methods have been proposed for treating phenylketonuria (PKU), which include administering to a subject in need of said treatment a composition comprising an mRNA encoding phenylalanine hydroxylase (PHA), in an effective dosage and with an effective administration interval, thereby reducing the intensity, severity, frequency, or delay of the onset of at least one symptom or symptom of PKU. In some embodiments of the invention, the mRNA is encapsulated in a liposome that contains one or more cationic lipids, one or more non-cationic lipids, one or more cholesterol-based lipids and one or more PEG-modified lipids.
